Complement-mediated vasoconstriction and graft rejection.

چکیده
The hyperacute rejection of renal allografts has much in common with pigto-dog renal xenograft rejections. An analysis of the xenograft model revealed new functional aspects of graft rejection that appear to be an integral part of the humoral immune injury. Anaphylatoxin or C5a, a cleavage product of the fifth complement component was shown to play a major role in the rejection of isolated kidneys; it was generated in pig and dog serum by activation of the complement system and caused severe vasoconstriction in the renal vasculature. It is suggested that histocompatibility antibodies and complement may affect allograft survival via this potent mediator.

CD4+ T cells and complement independently mediate graft ischemia in the rejection of mouse orthotopic tracheal transplants.
RATIONALE While microvascular injury is associated with chronic rejection, the cause of tissue ischemia during alloimmune injury is not yet elucidated. OBJECTIVE We investigated the contribution of T lymphocytes and complement to microvascular injury-associated ischemia during acute rejection of mouse tracheal transplants. METHODS AND RESULTS Using novel techniques to assess microvascular i...

Antibody-mediated rejection is well established for renal allografts but remains controversial for lung allografts. Cardinal features of antibody-mediated rejection in renal allografts include antibodies to donor human leukocyte antigen (HLA) and evidence for antibody action, such as complement activation demonstrated by C4d deposition. We report a lung allograft recipient with circulating anti...

Skin allografts survived longer on ALS-treated, complement-deficient (C5 negative) recipients than on ALS-treated, complement-competent (C5 positive) recipients. Administration of C5-positive serum to C5-negative, ALS-treated recipients resulted in reduced graft survival. A percentage of grafts from ALS-treated, C5-positive donors was rejected when transferred to untreated syngeneic recipients;...
متن کاملCutting Edge: NK cells mediate IgG1-dependent hyperacute rejection of xenografts.
Classic hyperacute rejection is dependent on the activation of the terminal components of complement. Recently, xenoantibodies with limited abilities to activate the classical pathway of complement in vitro have been implicated in the acute vascular rejection of xenografts. It is unclear how these Abs affect their pathogenic activities in vivo. In this study, we demonstrate the ability of an an...
متن کامل- 1 - Complement in Renal Transplant Rejection
The complement system plays multiple pathogenetic roles in rejection of renal allografts and one of the components, C4d, is an important diagnostic marker of antibody mediated rejection (1, 2). Complement proteins (e.g., C3) arose as an ancient, innate defense system in invertebrates that preceded the adaptive immune system in phylogeny. It is not surprising that during evolution these compleme...
